产品介绍The protein encoded by this gene is a transmembrane (type I) heparan sulfate proteoglycan and is a member of the syndecan proteoglycan family. The syndecans mediate cell binding, cell signaling, and cytoskeletal organization and syndecan receptors are required for internalization of the HIV-1 tat protein. The syndecan-1 protein functions as an integral membrane protein and participates in cell proliferation, cell migration and cell-matrix interactions via its receptor for extracellular matrix proteins. Altered syndecan-1 expression has been detected in several different tumor types. While several transcript variants may exist for this gene, the full-length natures of only two have been described to date. These two represent the major variants of this gene and encode the same protein. [provided by RefSeq, Jul 2008]

CD138（Syndecan-1，SDC1）是跨膜硫酸乙酰肝素蛋白聚糖（分子量约34kDa），核心功能与研究价值包括：

浆细胞标志物：在多发性骨髓瘤（MM） 中，CD138是肿瘤浆细胞的特异性表面抗原（阳性率>95%），是骨髓活检IHC和流式细胞术分型的核心指标；

肿瘤微环境调控：通过胞外域肝素 sulfate 链结合FGF、VEGF等生长因子，促进肿瘤血管生成；胞内域与细胞骨架蛋白（如肌动蛋白）相互作用，增强肿瘤细胞黏附与侵袭；

治疗靶点：CD138单抗偶联药物（如Indatuximab ravtansine）可靶向递送细胞毒素至MM细胞，临床前研究显示对耐药性MM有效。

实验操作关键要点

IHC-P多发性骨髓瘤诊断：

样本处理：骨髓活检石蜡切片，EDTA缓冲液（pH9.0）高压修复20分钟（优于柠檬酸盐缓冲液，保留硫酸肝素链抗原性）；

抗体孵育：一抗1:200稀释（5% BSA+TBST），4℃湿盒孵育过夜，HRP二抗1:500室温孵育30分钟；

阳性判读：骨髓瘤细胞细胞膜呈棕黄色强阳性，正常浆细胞弱阳性，淋巴细胞/粒细胞阴性（与CD38共染可提高诊断特异性）。

流式细胞术（MM细胞分选）：

样本制备：骨髓单个核细胞用PBS+2% FBS洗涤，CD138抗体（如艾普蒂生物APC标记）1:50稀释，4℃避光孵育30分钟；

抗体组合：与CD38-PE、CD45-FITC共染，MM细胞典型表型为CD138⁺CD38⁺⁺CD45⁻/dim；

注意事项：CD138易因蛋白酶降解脱落，样本需新鲜处理（离体后2小时内染色）。

IF肿瘤细胞黏附研究：

与整合素β1（ITGB1） 共染，CD138（绿色）与ITGB1（红色）在细胞边缘共定位，促进肿瘤细胞与细胞外基质黏附（共定位系数>0.5）。
