Astrophloxine is a fluorescent imaging probe. Astrophloxine targets antiparallel dimers. Astrophloxine can be used to detect aggregated Aβ in cerebrospinal fluid and brain tissue samples of Alzheimer's disease (AD) mice.

For CSF analyses: 1. 1 μL of CSF samples were treatedwith 1 μL of 10X protease inhibitors and diluted the mixture with 8 μL of binding buffer. 2. The dye was diluted to 0.5 μM with binding buffer. 3. The individual samples and dye was added to the wells of a 96-well half-area black microplate with a sample to dye ratio of 1:3. 4. The samples were distributed in triplicates and detected at λex=540 nm/λem=570 nm[1].

Aβ concentrations in CSF samples of WT and APP/PS1 TG mice were detected by adding 0.5 μM of Astrophloxine with a sample to dye ratio of 1:3. The results indicated a significant increase when comparing CSF samples of adult and aged APP/PS1 mice. Astrophloxine can stain insoluble Aβ plaques in 9-month-old female 5XFAD mouse brain cryosections[1].
