TD-0212 TFA is an orally active dual pharmacology antagonist of angiotensin II type 1 receptor (AT1) and inhibitor of neprilysin (NEP)(pKi of 8.9 for AT1 and a pIC50 of 9.2 for NEP).

TD-0212 provides the enhanced activity of dual AT1/NEP inhibition and may have a lower risk of angioedema than dual ACE/NEP inhibition.

In renin-dependent and non-dependent hypertension models, TD-0212 can lower blood pressure, similar to omapatrilat and the combination of AT1 receptor antagonist and NEP inhibitor.
