背景信息：

Clusterin (CLU, 载脂蛋白 J) 是一种多功能糖蛋白，在大多数组织中广泛表达。Clusterin 的可发挥分泌性伴侣蛋白的功能，与应激诱导蛋白相互作用并使其稳定，以防止其沉淀 (1,2)。研究显示 clusterin 在阿尔茨海默症中起到保护作用，clusterin 可以隔离 β 淀粉样蛋白 (1-40) 肽以形成长寿且稳定的复合体，从而阻止淀粉样蛋白原纤维的形成 (3-5)。
除分泌性蛋白外，还有几种胞内同工型均定位到胞核、线粒体、细胞浆和内质网上。这些同工型的亚细胞分布促使 clusterin 产生不同的功能。还有研究表明， clusterin 参与了膜的循环、细胞黏附、细胞增殖、凋亡和肿瘤存活 (6-9) 等活动。clusterin 前体在翻译后裂解为成熟 clusterin α 和 clusterin β。Clusterin α 和 β 通过形成二硫键来产生异二聚体 (10)。
