背景信息：
在淋巴细胞受体刺激后，Ras 信号转导通路激活在某种程度上会导致淋巴细胞激活。鸟嘌呤核苷酸交换因子 (GEF) RasGRP 家族能在 Ras 家族小 GTP 酶上催化 GDP 交换为 GTP，从而促进形成活化的 GTP 结合形式。二脂酰甘油 (DAG) 或佛波酯结合 RasGRP 家族成员会导致转位到细胞膜并刺激其活性 (1,2)。T 细胞表达 RasGRP1，而 B 细胞则表达 RasGRP1 和 RasGRP3。RasGRP3 在 B 细胞受体 (BCR) 激活信号偶联到 Ras 信号转导的过程中非常重要 (3)。在 BCR 的刺激下，RasGRP3 在 Thr133 位点 被 PKC 磷酸化。在 DAG 的刺激下，这种磷酸化活动会进一步激活 RasGRP3，从而刺激 PKC 活性 (4,5)。
