L-168,049 (L 168,049; L168,049) is a novel, selective, orally bioactive and non-peptidyl antagonist of human glucagon receptor (GR) with anti-diabetic activity. It activates human GR with an IC50 value of 3.7 nM, but exhibits moderate affinity to murine and canine GRs. It inhibits glucagon-stimulated cAMP synthesis in murine liver membranes and in CHO cells expressing hGR.
