生物活性
Saracatinib (AZD0530)是一种有效的Src抑制剂,IC50为2.7 nM,对c-Yes, Fyn, Lyn, Blk, Fgr和Lck等也有活性;但对Abl和EGFR (L858R和L861Q)活性较低。Saracatinib也有效抑制其他Src酪氨酸激酶家族成员,包括c-Yes, Fyn, Lyn, Blk, Fgr, 和Lck,IC50为4到10 nM。Saracatinib有效抑制SrcY530F突变的NIH 3T3细胞,IC50为80 nM。在NBT-II膀胱癌细胞中,Saracatinib显著阻断HT1080细胞通过立体骨胶原基质的入侵,且完全抑制EGF诱导的细胞分散。

方法 | Cell proliferation was assessed using a colorimetric 5-bromo- 20 -deoxyuridine (BrdU) Cell Proliferation ELISA kit (Roche Diagnostics GmbH), as described previously (Ple' et al., 2004). Briefly, cells were plated onto 96-well plates (1.5*104 cells/ well), the following day 0.039–20 μM AZD0530 in DMSO (at a final concentration of 0.5%) was added and the cells were incubated for 24 h. The cells were pulse labeled with BrdU for 2 h and fixed. Cellular DNA was then denatured with the provided solution and incubated with antiBrdU peroxidase for 90 min. Following three washes with phosphate-buffered saline, tetramethylbenzidine substrate solution was added and the plates were incubated on a plate shaker for 10–30 min until the positive control absorbance at 690 nm was approximately 1.5 absorbance units. |
