5-HT7受体在调节睡眠和昼夜节律中发挥作用.此受体通过中枢和外周机制,也在其他CNS疾病中发挥一定的作用,包括认知障碍\焦虑和偏头痛[2].
通过5-HT7受体传递信号,5-羟色胺为增强T细胞活化提供辅助信号.相比于盐处理的对照,PCPA处理的小鼠中T细胞增殖受到显著抑制.PCPA的抑制效应约为18%-49%.加入1 μM 外源性5-HT能够完全恢复T细胞增殖.类似于外源性5-HT,加入1 μM AS 19也能于48小时后完全恢复T细胞增殖能力[4].
SB-258719是选择性的5-HT7受体拮抗剂.10 mg/kg的AS-19与同剂量的SB-258719协同处理(对其本身无效应),在热板和甩尾实验中,AS-19的抗痛觉过敏效应皆被逆转.表明AS-19通过5-HT7受体发挥抗痛觉过敏效应[3].SB269970也是选择性5-HT7受体拮抗剂.一些大鼠通过第二鞘内导管用7 μl 5 mM的SB269970预处理,在所有时间节点,AS-19诱导的PMF被SB269970的预处理所废除,表明AS-19通过5-HT7受体的活化而产生PMF[5].
