OG-L002 is a recently developed inhibitor with IC50of 20 nM at LSD1 and much lower activity at MAO-A and MAO-B (1.38 and 0.72 μM, respectively). OG-L002 exhibits potent anti-viral activityin vitroand in mouse models of HSV infection. Lysine specific demethylase 1 (LSD1) is a histone demethylase that removes methyl groups from lysine 4 or 9 of H3 histone tails. Inhibition of LSD1 leds to suppression of herpes simplex and herpes zoster viral infections and viral reactivation from latency. MAO inhibitors (pargyline, tranylcypromine) are known to inhibit LSD1, but with low potency and selectivity. OG-L002 is a more selective and potent tool for LSD1 inhibition.
